There are a few routes I'd like to follow from one end to the other, Rte. 66 being one of them. Two others that come to mind immediately are U.S. 59 and U.S. 6.
While I'm way too young to understand the nostalgia of 66 (they had probably long since taken down the construction signs for I-40 by the time I was born), I think I can appreciate the significance of it.
I haven't really been on either 6 or 59, but I'm sure they have their own allure.
